目的 探讨Clusterin在细胞外水平抗膀胱癌细胞的自然死亡作用。 方法 应用流式细胞术 (FCM )、MTT试验和伊红染色等方法 ,通过检测不同浓度Clusterin对膀胱癌EJ细胞的自然死亡率及丝裂霉素杀伤后的膀胱癌细胞死亡率的影响 ,分析Clusterin对EJ细胞的抗死亡作用。 结果 Clusterin可以明显降低膀胱癌细胞的自然死亡率 ,还可以对抗丝裂霉素对EJ细胞的杀伤作用。 结论 检测Clusterin的表达特性将有助于临床对膀胱癌的耐药性及用药后复发做出评估分析 ,为应用Clusterin抗体治疗膀胱癌提供理论依据。
Objective To investigate the effect of Clusterin on the natural death of bladder cancer cells at extracellular level. Methods FCM, MTT assay and eosin staining were used to detect the effects of different concentrations of Clusterin on the natural death rate of bladder cancer cells and the mortality of bladder cancer cells after mitomycin C treatment. Clusterin on EJ cells anti-death effect. Results Clusterin can significantly reduce the natural death rate of bladder cancer cells, but also against the killing effect of mitomycin on EJ cells. Conclusion The detection of Clusterin expression characteristics will help clinical evaluation of bladder cancer drug resistance and relapse after treatment to provide a theoretical basis for the application of Clusterin antibody in the treatment of bladder cancer.
