目的:观察芝麻素与维生素E联用对代谢综合征大鼠心肌损伤的保护作用。方法:高脂高糖诱导大鼠代谢综合征24周,第9周起每天口服1次芝麻素(30 mg/kg)、芝麻素+维生素E(30+20、15+20 mg/kg)和维生素E(20mg/kg)含药饲料,共16周。于末次给药后,称体重(BW)、全心(HWW)和左心室湿重(LVWW);测血糖、血脂和血压、心肌组织羟脯氨酸(HYP)、过氧化氢(H2O2)、一氧化氮(NO)含量及超氧化物歧化酶(SOD)和过氧化氢酶(CAT)活性;HE染色观察心肌组织病理变化;免疫组化法表达心肌诱导型一氧化氮合酶(iNOS)和硝基酪氨酸(NT)含量。结果:与模型组相比,芝麻素+维生素E(30+20 mg/kg)组血糖、血脂和血压明显降低,BW、HWW和LVWW显著下降;心肌H2O2、HYP和NO含量明显减少;SOD和CAT活性明显升高;心肌iNOS表达和NT含量减少;病理损伤明显减轻。上述结果优于单用芝麻素组或维生素E组(P<0.01或P<0.05)。结论:芝麻素(30mg/kg)与维生素E(20mg/kg)联用具有协同抗氧化和保护代谢综合征大鼠心肌的作用。
Objective: To observe the protective effects of sesamin and vitamin E on myocardial injury in rats with metabolic syndrome. Methods: Metabolic syndrome was induced in rats with high fat and high glucose for 24 weeks. From the 9th week, sepsin (30 mg / kg), sesamin + vitamin E (30 + 20,15 + 20 mg / kg) Vitamin E (20mg / kg) containing medicated feed, a total of 16 weeks. Body weight (BW), total heart (HWW) and left ventricular wet weight (LVWW) were measured after the last administration. Blood glucose, blood lipid and blood pressure, myocardial hydroxyproline (HYP), hydrogen peroxide (NO) and the activities of superoxide dismutase (SOD) and catalase (CAT). The pathological changes of myocardium were observed by HE staining. The expression of iNOS was detected by immunohistochemistry. And nitrotyrosine (NT) content. Results: Compared with the model group, the levels of blood glucose, blood lipids and blood pressure were significantly lower in sesamin + vitamin E (30 + 20 mg / kg) groups and the levels of BW, HWW and LVWW were significantly decreased; the contents of H2O2, CAT activity was significantly increased; myocardial iNOS expression and NT content decreased; pathological injury significantly reduced. The above results were better than those with sesamin alone or vitamin E (P <0.01 or P <0.05). Conclusion: The combination of sesamin (30mg / kg) and vitamin E (20mg / kg) has synergistic anti-oxidation and protective effect on the myocardium of rats with metabolic syndrome.
