目的:建立柱前衍生化HPLC检测不同基原珍珠粉及贝壳粉中7种主要氨基酸的方法。方法:以异硫氰酸苯酯为柱前衍生化试剂,采用C18色谱柱,流动相A:0.1 mol/L醋酸钠溶液(用醋酸调p H值至6.5),流动相B:乙腈,梯度洗脱,检测波长254 nm。结果:氨基酸衍生溶液在24 h内保持稳定,7种氨基酸线性相关系数均大于0.9995,加样回收率在93.16%~103.31%之间。结论:本方法分离效果好,精密度和重复性良好,可用于珍珠粉及贝壳粉中7种水解氨基酸含量的测定。
OBJECTIVE: To establish a method for the determination of seven major amino acids in pearl and shell powder of different bases by precolumn derivatization. Methods: Phenylisothiocyanate was used as pre-column derivatization reagent. The mobile phase A was consisted of 0.1 mol / L sodium acetate solution (p H value adjusted to 6.5 with acetic acid), mobile phase B: acetonitrile, gradient Elution, detection wavelength 254 nm. Results: The amino acid derivative solution remained stable within 24 h. The linear correlation coefficients of seven amino acids were all greater than 0.9995. The recoveries of the two amino acids ranged from 93.16% to 103.31%. Conclusion: The method has good separation effect, good precision and repeatability, and can be used for the determination of seven kinds of hydrolyzed amino acids in pearl powder and shell powder.
