目的:研究脑源性神经营养因子(BDNF)与脑缺血严重程度的关系。方法:制作大鼠大脑中动脉阻塞(MCAO)模型,于30min MCAO及100min MCAO后再灌注4h、8h、24h后取大鼠脑组织进行BDNF免疫组织化学染色,观察缺血脑皮层内BDNF阳性细胞所占比例,并与假手术对照组比较。结果:30min MCAO组再灌注4h、8h、24h与假手术对照组比较缺血脑皮层BDNF阳性细胞所占比例无明显差异(P>0.05);100min MCAO再灌注4h、8h、24h组与假手术对照组比较缺血脑皮层BDNF阳性细胞所占比例增加,差异有统计学意义(P<0.05)。结论:脑组织缺血可导致缺血神经元BDNF表达明显增加。
Objective: To study the relationship between brain derived neurotrophic factor (BDNF) and the severity of cerebral ischemia. Methods: The middle cerebral artery occlusion (MCAO) model was made in rats. After 30 min MCAO and 100 min MCAO reperfusion for 4 h, 8 h and 24 h, the brain tissue of rats were taken for BDNF immunohistochemical staining to observe the expression of BDNF positive cells Proportion and compared with the sham operation control group. Results: There was no significant difference in the percentage of BDNF positive cells in ischemic cerebral cortex between MCAO group and control group at 4h, 8h, 24h after reperfusion for 30min (P> 0.05) The proportion of BDNF positive cells in ischemic cortex increased in the control group, the difference was statistically significant (P <0.05). CONCLUSION: Cerebral ischemia can result in marked increase of BDNF expression in ischemic neurons.
