目的探讨肺结核患者的血清中血管内皮生长因子(VEGF)、白介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)的表达与变化。方法对疑似肺结核病患者采样痰涂片抗酸染色法和酶联免疫反应法检测筛选检测,在确诊为肺结核患者时,静脉采血,通过Real-time RT-PCR法和Western-blot检测其血清中VEGF、TNF-α、IL-6 mRNA和蛋白表达量的变化与正常健康人进行比较。结果通过对95例疑似结核患者筛选,确诊有63例肺结核患者,通过Real-time RT-PCR检测发现VEGF、TNF-α、IL-6 mRNA的表达量明显著高于正常健康人组,Western-blot检测结果表明肺结核患者体内VEGF、TNF-α、IL-6蛋白表达量显著高于对照组。结论结核患者体内VEGF、IL-6表达水平升高,通过检测研究VEGF、TNF-α、IL-6表达水平的变化可研究结核杆菌对人体致病机制。
Objective To investigate the expression of vascular endothelial growth factor (VEGF), interleukin-6 (IL-6) and tumor necrosis factor-α (TNF-α) in patients with pulmonary tuberculosis. Methods The sputum smear acid-fast staining and enzyme-linked immunosorbent assay (ELISA) were used to screen and detect the suspected pulmonary tuberculosis patients. When diagnosed as pulmonary tuberculosis patients, blood samples were taken from venous blood and detected by Real-time RT-PCR and Western-blot The changes of VEGF, TNF-α, IL-6 mRNA and protein expression were compared with those in normal healthy subjects. Results Ninety-five patients with tuberculosis were screened from 95 patients with suspected tuberculosis. The expression of VEGF, TNF-α and IL-6 mRNA was significantly higher than that of normal healthy people by Real-time RT-PCR. Western- The results of blot showed that the expression of VEGF, TNF-α and IL-6 in tuberculosis patients was significantly higher than that in control group. Conclusion The expression of VEGF and IL-6 in tuberculosis patients is elevated. The pathogenic mechanism of Mycobacterium tuberculosis to human body can be studied by examining the changes of the expression of VEGF, TNF-α and IL-6.
