目的:为了解感染日本血吸虫后小鼠不同脏器的免疫反应,我们观察了小鼠肝、脾、小肠Th2因子的变化并探讨其意义.方法:采用ABC免疫组化法,并用多媒体病理图文定量分析,动态研究感染后8,10,12周IL-4、IL-5、IL-10的变化.结果:在感染小鼠肝脏IL-4明显高于正常对照(P<0.01).并随感染时间的延长而逐步升高,IL-5和IL-10亦有相同变化,但IL-4最高,IL-10次之,IL-5居后;在感染小鼠的脾脏IL-4、IL-5、IL-10亦随感染时间的延长逐步缓慢升高,但IL-4在感染的第10周,IL-5在感染的第12周,IL-10从感染的第8周起与正常对照比较才有显著意义(P<0.05);在感染小鼠的小肠IL-4、IL-5、IL-10随感染时间延长呈缓慢升高趋势,从感染的第8周起与正常比较有显著差异(P<0.05~0.01).结论:感染日本血吸虫病的小鼠,肝脏是Th2因子免疫应答的主要器官,IL-4、IL-5和IL-10呈同步平行变化,以IL-4升高最明显;脾脏是调节免疫的器官,上述Th2因子的变化明显低于肝脏;而感染小鼠的小肠亦不是免疫应答的主要场所,其Th2因子的升高可能是整体反应的一部分.
OBJECTIVE: To investigate the immune response of different organs of mice infected with Schistosoma japonicum, we observed the changes of Th2 factor in mice liver, spleen and small intestine and discussed the significance. Methods: Immunohistochemical ABC method was used to quantitatively analyze the levels of IL-4, IL-5 and IL-10 at 8, 10 and 12 weeks after infection with quantitative analysis of pathological images. Results: IL-4 in the liver of infected mice was significantly higher than that of the normal controls (P <0.01). IL-5 and IL-10 also had the same changes, but IL-4 was the highest, IL-10 was the second, and IL-5 was later; in the infected mice, IL-4 , IL-5 and IL-10 gradually increased gradually with the prolongation of infection time, but IL-4 in the 10th week of infection, IL-5 in the 12th week of infection, IL-10 from the 8th week of infection (P <0.05). IL-4, IL-5 and IL-10 in small intestine of infected mice increased slowly with the prolongation of infection time. From the 8th week of infection Compared with the normal significant difference (P <0.05 ~ 0.01). Conclusion: The liver of mice infected with schistosomiasis japonica is the main organ of immune response to Th2 factor. The IL-4, IL-5 and IL-10 showed synchronous and parallel changes, with the most obvious increase of IL-4; Organs, the change of Th2 factor was obviously lower than that of the liver. However, the small intestine of infected mice was not the main site of immune response. The increase of Th2 factor may be part of the overall response.
