目的:构建肾纤维化来源人肾间质FBS的离体培养模型。方法:用慢性梗阻性肾病(UUO)患者手术取材标本,体外培养5代细胞,观察细胞的形态变化和生长状况,并用免疫细胞化学方法对其进行鉴定。结果:培养的细胞呈柳叶形,核1~2个;第2~4代细胞在培养过程中生长速度最快;免疫细胞化学鉴定表达波形蛋白(vimen -tin) ;结蛋白(desmin)和角蛋白(keratin)表达为阴性。结论:在体外成功培养出纤维化来源的肾间质成纤维细胞,第2~4代体外培养细胞是细胞试验的最佳选择。
Objective: To construct an in vitro culture model of human renal interstitial fibrosis FBS. Methods: The specimens of patients with chronic obstructive nephropathy (UUO) were collected by surgery. The cells were cultured in vitro for 5 generations. Morphological changes and growth of the cells were observed. Immunocytochemistry was used to identify the cells. Results: The cultured cells were willow-shaped with 1-2 nuclei. The second to fourth passage cells grew fastest during the culture process. Immunocytochemistry identified vimen -tin, desmin and Keratin expression is negative. CONCLUSIONS: Fibroblast-derived renal interstitial fibroblasts were successfully cultured in vitro. The second to fourth generation of cultured cells in vitro were the best choice for cell assay.
