目的:运用近红外漫反射光谱技术(NIRS)结合偏最小二乘法(PLS)建立野菊花药材中蒙花苷含量的快速测定方法。方法:采集野菊花药材的近红外漫反射光谱,采用HPLC测定野菊花药材中蒙花苷含量作为参考值,将近红外光谱与蒙花苷含量参考值进行关联,建立野菊花药材中蒙花苷含量的定量预测模型,并对模型进行验证。结果:所建立的定量分析模型的内部交叉验证决定系数(R2)为0.999 09,校正均方根偏差(RMSEC)为0.013 6,内部交叉验证均方差(RMSECV)为0.039 70,验证集样品预测相关系数(r)为0.997 6,预测均方偏差(RMSEP)为0.018 5。结论:近红外光谱法操作简便,测定快速准确,可以用于河南产野菊花药材中蒙花苷含量的快速测定。
Objective: To establish a rapid method for the determination of monatin content in wild chrysanthemum by near infrared diffuse reflectance spectroscopy (NIRS) combined with partial least squares (PLS). Methods: The near-infrared diffuse reflectance spectra of wild chrysanthemum medicinal materials were collected. HPLC was used to determine the content of monatin in wild chrysanthemum medicinal materials as a reference value. The correlation between near-infrared spectroscopy and reference value of monatin was established. Quantitative prediction model, and verify the model. Results: The internal cross validation coefficient (R2) was 0.999 09, the RMSEC was 0.013 6, and the RMSECV was 0.039 70. The validation set was correlated with sample prediction The coefficient (r) was 0.997 6, and the prediction mean square deviation (RMSEP) was 0.018 5. Conclusion: Near infrared spectroscopy is simple, rapid and accurate, and can be used for the rapid determination of the content of monolin in Henan wild chrysanthemum medicinal materials.
