Courtenary软琼脂集落试验已被广泛运用于研究人肿瘤标本对放疗及化疗的敏感性。作者用其测定37例卵巢瘤、6例子宫颈癌、5例乳腺癌、4例膀胱癌、4例头颈上皮细胞癌、2例精原细胞瘤及8例黑色素瘤共7种肿瘤细胞的放射存活曲线,探讨细胞体外放射敏感性和临床放射可治愈性之间可能存在的关系。手术取得癌组织即置4℃培养液中,经机械离散及酶处理等,获单细胞悬液,用Siemens Stabili pa X线机,220Kv、19—20mA,经0.5mm铜滤过照射细胞悬液,剂量为3.0Gy/分。以Courtenary软琼脂集落试验测定细胞的存活曲线及有关参数。实验结果表明,同一组织类型的各个肿瘤间的2.0Gy存活率和存活曲线是很不同的,而存活曲线参数(Do,Dq及n)与2.0Gy存活率在上述7种肿瘤间却没有显著的统计学差异。这些观察结果提示,直接
Courtenary soft agar colony assay has been widely used to study the sensitivity of human tumor specimens to radiotherapy and chemotherapy.
