研究微波辅助提取,反相高效液相色谱测定花菜中VC含量的方法。花菜匀浆后,以1%草酸为萃取溶剂,微波提取、净化后,采用恒流的方法进行液相色谱定量检测,以0.1%草酸溶液为流动相,流速1.0 mL/min,采用C18反相色谱柱分离,在254 nm波长下检测。结果表明,VC质量浓度在0.02~0.12 mg/mL范围内线性良好,相关系数为0.999 6,检测限0.02 mg/kg,回收率在100.5%~104.6%之间,相对标准偏差为1.20%。该方法能有效提取蔬菜中VC,测定结果稳定,重现性好,可以用于蔬菜中VC的检测。
Study on Microwave Assisted Extraction and RP - HPLC Determination of VC in Cauliflower. After the cauliflower was homogenized, 1% oxalic acid was used as the extraction solvent and extracted by microwave. After purification, the sample was detected by constant current method. The mobile phase consisted of 0.1% oxalic acid solution with a flow rate of 1.0 mL / min and C18 reversed phase Column separation, detection at 254 nm wavelength. The results showed that the linearity of VC ranged from 0.02 to 0.12 mg / mL. The correlation coefficient was 0.999 6. The detection limit was 0.02 mg / kg. The recoveries ranged from 100.5% to 104.6%. The relative standard deviations were 1.20%. The method can effectively extract VC from vegetables, has stable measurement results and good reproducibility, and can be used for detecting VC in vegetables.
