成年大鼠脑缺血诱导新生神经元形成纹状体-黒质投射的研究

  为了研究在成年动物脑内缺血诱导的纹状体新生神经元是否能在结构上整合到现有的神经环路中,在动物缺血再灌注第1和11周将荧光金(一种逆向示踪剂)注射到黑质网状部,我们通过标记BrdU并结合免疫组化的方法标记新生神经元,同时应用激光共聚焦扫描的方法研究纹状体内新生神经元的投射情况。结果显示,正常或缺血再灌注的动物脑内均可在同侧检测到FG阳性信号。并且,在缺血再灌注第12周缺血侧纹状体的新生神经元上检测到FG的表达(BrdU+-FG+双标细胞),而缺血再灌注2周却没有检测到双标细胞。这提示在成年动物脑缺血后纹状体新生神经元可以形成长投射到黒质网状部,也意味着了这些新生神经元具有逆向轴浆转运的功能。形态学的结果表明,纹状体作为一个非神经发生区,缺血后的新生神经元可以建立起从纹状体到黒质网状部的神经环路。这部分的结果对于研究缺血性脑中风后的神经功能恢复将具有十分重要的意义。
