常用中药提取物对NIH3T3细胞生长影响

  目的 高通量筛选技术使天然产物的大规模系统活性研究成为可能,可以达到快速准确了解中药活性组分及药理作用的目的.通过样品对正常细胞的毒性作用评价可以帮助预测动物毒性作用,同时在药物开发早期去除可能的毒性成分.本研究利用本课题组前期建立的天然产物提取物样品库中的样品,结合高通量筛选技术评价了常用中药单味药提取物成分对小鼠成纤维细胞系NIH3T3生长的影响,以其获得常用中药对正常细胞的细胞毒作用结果.方法 将中药单味药分别采用石油醚,乙醇,水三种提取方法获得相应提取物,共获得573个样品.将NIH-3T3细胞以3000每孔密度接种于96孔细胞培养板中,细胞培养液为180μl含10% FBS的RPM1640培养基.次日细胞长成单质后,加入20μl的待测药物,37℃5% CO2条件下继续培养36 h.MTT测定细胞活力,以生理盐水作为空白对照.相对于对照孔的细胞数作为细胞增殖与抑制的比率.结果 14.5%样品对细胞增殖的抑制作用达到50%以上,而抑制率大于80%的样品占4.5%.其中抑制率大于90%的样品包括:竹黄乙醇提取物,三条筋石油醚提取物;山楂水提取物;三台红花乙醇提取物;粉葛石油醚提取物;寄生石油醚提取物;草果药水提取物;苏木水提取物;金花小孽乙醇提取物;木贼草乙醇提取物等.结论 中药提取物的成分比较复杂,多种提取物具有抑制正常细胞生长的作用,在研究中药提取物的药理作用时要注意其可能产生的细胞毒作用影响.
